CD47 expression is decreased in hematopoietic progenitor cells in patients with myelofibrosis BJMBR
Nonino,A.; Nascimento,J.M.; Mascarenhas,C.C.; Mazzeu,J.F.; Pereira,R.W.; Jacomo,R.H..
Myelofibrosis (MF) is characterized by increased circulating hematopoietic progenitor cells (HPCs), abnormal cytokine levels, and the survival advantage of neoplastic progenitors over their normal counterparts, which leads to progressive disappearance of polyclonal hematopoiesis. CD47 is a surface glycoprotein with many functions, such as acting as a phagocytosis inhibitor of the expressing cell, that is increased in normal hematopoietic stem and progenitor cells mobilized into the blood and several human cancer-initiating cells, such as in acute myeloid leukemia. We compared CD47 expression in hematopoietic stem and progenitor cells of patients with MF and controls and found it to be decreased in progenitors of MF. Exposure of control HPCs to the...

Humoral and cellular immune response of mice challenged with Yersinia pestis antigenic preparations BJID
Leal,Elida A.; Moreira,Josimar D.; Nunes,Fernanda F.; Souza,Larissa R.; Martins,Janaina M.; Toledo,Vicente P.C.; Almeida,Alzira M.P.; Guimarães,Tania M.P..
ABSTRACT Objectives: The plague, which is an infectious disease caused by Yersinia pestis, still threatens many populations in several countries. The worldwide increase in human plague cases and the potential use of the bacteria as a biological weapon reinforce the need to study the immunity that is induced by potential vaccine candidates. To determine the immunogenicity of antigenic preparations based on the F1 protein and the total extract from Y. pestis, we assessed the role of these antigens in inducing an immune response. Methods: The immunogenicity of antigenic preparations based on the Y. pestis (YP) total extract and the Y. pestis fraction 1 capsular antigen protein (F1) was determined in Swiss-Webster mice immunized with 40 µg or 20 µg for each...

Humoral response to low molecular weight antigens of Mycobacterium tuberculosis by tuberculosis patients and contacts BJMBR
Beck,S.T.; Leite,O.M.; Arruda,R.S.; Ferreira,A.W..
Much effort has been devoted to the identification of immunologically important antigens of Mycobacterium tuberculosis and to the combination of target antigens to which antibodies from serum of tuberculous patients could react specifically. We searched for IgG antibodies specific for antigens of 45 to 6 kDa obtained after sonication of the well-characterized wild M. tuberculosis strain in order to detect differences in the antibody response to low molecular weight antigens from M. tuberculosis between patients with pulmonary tuberculosis and contacts. Specific IgG antibodies for these antigens were detected by Western blot analysis of 153 serum samples collected from 51 patients with confirmed pulmonary tuberculosis. Three samples were collected from each...

Identification of Mycobacterium bovis antigens by analysis of bovine T-cell responses after infection with a virulent strain BJMBR
Alito,A.; McNair,J.; Girvin,R.M.; Zumarraga,M.; Bigi,F.; Pollock,J.M.; Cataldi,A..
Purification and characterization of individual antigenic proteins are essential for the understanding of the pathogenic mechanisms of mycobacteria and the immune response against them. In the present study, we used anion-exchange chromatography to fractionate cell extracts and culture supernatant proteins from Mycobacterium bovis to identify T-cell-stimulating antigens. These fractions were incubated with peripheral blood mononuclear cells (PBMC) from M. bovis-infected cattle in lymphoproliferation assays. This procedure does not denature proteins and permits the testing of mixtures of potential antigens that could be later identified. We characterized protein fractions with high stimulation indices from both culture supernatants and cell extracts....

Identification of putative new Escherichia coli flagellar antigens from human origin using serology, PCR-RFlP and DNA sequencing methods BJID
Tiba,Monique Ribeiro; Moura,Claúdia de; Carazzolle,Marcelo Falsarella; Leite,Domingos da Silva.
Escherichia coli has been isolated frequently, showing flagellar antigens that are not recognized by any of the 53 antisera, provided by the most important reference center of E. coli, The International Escherichia and Klebsiella Center (WHO) of the Statens Serum Institute, Copenhagen, Denmark. The objective of this study was to characterize flagellar antigens of E. coli that express non-typeable H antigens. The methods used were serology, PCR-RFLP and DNA sequencing. This characterization was performed by gene amplification of the fliC (flagellin protein) by polymerase chain reaction in all 53 standards E.coli strains for the H antigens and 20 E. coli strains for which the H antigen was untypeable. The amplicons were digested by restriction enzymes, and...

Main features of DNA-based immunization vectors BJMBR
Azevedo,V.; Levitus,G.; Miyoshi,A.; Cândido,A.L.; Goes,A.M.; Oliveira,S.C..
DNA-based immunization has initiated a new era of vaccine research. One of the main goals of gene vaccine development is the control of the levels of expression in vivo for efficient immunization. Modifying the vector to modulate expression or immunogenicity is of critical importance for the improvement of DNA vaccines. The most frequently used vectors for genetic immunization are plasmids. In this article, we review some of the main elements relevant to their design such as strong promoter/enhancer region, introns, genes encoding antigens of interest from the pathogen (how to choose and modify them), polyadenylation termination sequence, origin of replication for plasmid production in Escherichia coli, antibiotic resistance gene as selectable marker,...

Production of Trichophyton mentagrophytes antigens and their characterization in mice J. Venom. Anim. Toxins incl. Trop. Dis.
Venturini,J; Rossi-Ferreira,R; Arruda,MSP.
The participation of dermatophytic antigens in the host-parasite balance is still poorly understood. One of the difficulties encountered by researchers is the lack of dominant and specific antigens that can be used in such studies. In order to contribute to a better understanding of this aspect of infection, the present study identifies antigen fractions obtained from exoantigen and cytoplasmic extracts of Trichophyton mentagrophytes. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) revealed the presence of 13 proteins in the exoantigen extract, whose molecular weight ranged from 12.5 to 90 kDa. The cytoplasmic extract contained 18 protein fractions ranging from 11 to 110 kDa. Immunoblotting showed the presence of immunodominant...

Situação atual das estratégias para desenvolvimento de antígenos para o controle da neosporose em bovinos. Infoteca-e
SILVA, R. A. e; PIRAINE, R. E. A.; SANTOS JUNIOR, A. G. dos; CUNHA, R. C.; GONÇALVES, V. S.; SANTOS, F. D. S.; LEITE, F. P. L..

Sporothrix schenckii and sporotrichosis Anais da ABC (AABC)
Lopes-Bezerra,Leila M.; Schubach,Armando; Costa,Rosane O..
For a long time sporotrichosis has been regarded to have a low incidence in Brazil; however, recent studies demonstrate that not only the number of reported cases but also the incidence of more severe or atypical clinical forms of the disease are increasing. Recent data indicate that these more severe forms occur in about 10% of patients with confirmed diagnosis. The less frequent forms, mainly osteoarticular sporotrichosis, might be associated both with patient immunodepression and zoonotic transmission of the disease. The extracutaneous form and the atypical forms are a challenge to a newly developed serological test, introduced as an auxiliary tool for the diagnosis of unusual clinical forms of sporotrichosis.
